Human mitotic chromosomes consist predominantly of irregularly folded nucleosome fibres without a 30-nm chromatin structure.
How a long strand of genomic DNA is compacted into a mitotic chromosome remains one of the basic questions in biology. The nucleosome fibre, in which DNA is wrapped around core histones, has long been assumed to be folded into a 30-nm chromatin fibre and further hierarchical regular structures to form mitotic chromosomes, although the actual existence of these regular structures is controversia...
متن کاملHuman chromosomes. Evidence for autosomal sexual dimorphism.
The karyotypes of 100 males and 100 females, each assembled by the trypsin banding method, are examined in a study designed to investigate sex differences among autosomes. It is shown that female autosomes are consistently longer than those of the males, with respect to both the short and long arm measurements. In addition, discriminant analysis is used to distinguish between the male and femal...
متن کاملIsolation of folded chromosomes from Mycoplasma hyorhinis.
Folded chromosomes were isolated from Mycoplasma hyorhinis. When examined by electron microscopy, these molecules show variability of loop size, number of loops, total contour length and degree of twisting of the DNA. Sedimentation velocity was unaltered after treatment with RNase, proteinase K, SDS, temperatures up to 65 degrees C and NaCl concentrations from 0.1 M to 4 M.
متن کاملVisualization of G1 chromosomes: a folded, twisted, supercoiled chromonema model of interphase chromatid structure
We have used light microscopy and serial thin-section electron microscopy to visualize intermediates of chromosome decondensation during G1 progression in synchronized CHO cells. In early G1, tightly coiled 100-130-nm "chromonema" fibers are visualized within partially decondensed chromatin masses. Progression from early to middle G1 is accompanied by a progressive uncoiling and straightening o...
متن کاملHigher-order structure of human mitotic chromosomes.
From observations on the partial disintegration of isolated human metaphase chromosomes we propose that human metaphase chromatids have a rather simple organization based on the folding and coiling of a long, regular, hollow cylindrical structure with a diameter of about 4000 A. This cylindrical structure, the unit fiber, is postulated to be a super-solenoid formed by the coiling of a 300 A sol...
